Inducible Activity of Deleted Nopaline Syntheas Promoters by Insertion of Synthetic Oligomers in Transgenic Tobacco (Nicotiana tabacum)

|
The nopaline synthase (nos) promoter is regulated by various developmental and environmental factors. The nos control region essential for this response was studied by insertion of synthetic oligomers into deletion mutants using transient and stable transformation systems. 5¢¥and 3¢¥ deletion mutants were linked to form a set of internal deletion mutants and transient and stable assay showed that the region between nos promoter -131 and -112 was essential for nos promoter activity. Insertion of synthetic oligomers which were deleted at downstream hexamer motif reduced the promoter activity significantly. Insertion of 20 nucleotide sequences containing upstream and downstream hexamer motifs and a spacer region into nos deletion mutants altered the strength of expression of nos promoter. These results suggest that nos 20 element is essential for nos promoter to function. Insertion of nos 20 element into nos -101 mutant restored not only nos promoter activity but also responses to salicylic acid, methyl jasmonate, H©üO©ü, ascorbic acid and dithiothreitol.
